Internship on Decapod Project

Diponegoro Marine Biodiversity Project (DMBP) is a collaborative project between Yayasan Biodiversitas Indonesia (BIONESIA) and Diponegoro University, focusing on marine biodiversity research. This project located at  Terpadu Laboratory, Diponegoro University. One of the aims of this project is to perform capacity building activities, including giving training to the university students.This training was done as the internship program.

The internship students for this year were coming from 2 universities, including Trunojoyo University, Madura (Ahmad Rudianto, David Lolodo, and Desi Caturini Puspitasari) and Udayana University, Bali (Enex Yuniarti Ningsih and Shella Ayu Ardiana). The internship program was usually held for one month for every batch.

The first week activity for the internship students was introduction on the basic knowledge of genetics and how to perform laboratory work. The students were introduce to DNA Barcoding, basic molecular genetic and basic phylogenetic. For the lab-work activities, the students were trained to do DNA extraction, PCR (Polymerase Chain Reaction) and electrophoresis. The students were also given a chance to do the sequence editing and build phylogenetic tree from the sample’s sequences. At the end of the internship program, the students were encouraged to have a presentation about their activities in the lab.

One of the applications of DNA Barcoding method is on decapod identification, which was collected from various places around Indonesia. All of these samples were used in internship training. Decapod is usually difficult to identify due to its similar morphological characters and small size (cryptic species). Genetic was used as a tool to identify species of decapod collected, through the variation found in their DNA. The data of this result was expected to contribute to the genetic data worldwide.

Through this program, we hope that the internship student can give more contribution to Indonesian science, especially in the field of genetic and increase the understanding of young scientist on Indonesian marine biodiversity. Rudi, the intership student from Trunojoyo University, said that trough this training, he can increase his knowledge and skill in molecular genetic and apply it for his future research.
